The full-length cDNA encodes an 81-amino acid protein that has a calculated molecular mass of 9.2 kD and shares 72% identity with ATP6V0E1. Like ATP6V0E1, it contains 2 transmembrane domains.The ATP6V0E2 splice variant lacks exon 3, altering the reading frame and resulting in a 164-amino acid protein. RT-PCR showed that full-length ATP6V0E2 was expressed at high levels only in heart, brain, and kidney, although low expression was present in several other tissues. Expression of the ATP6V0E2 splice variant was restricted to heart, kidney, placenta, pancreas, and brain. Full-length ATP6V0E2 was detected in all brain regions examined, and the ATP6V0E2 splice variant was detected in all brain regions except frontal cortex.
