Glyoxalase Domain Containing Protein 5 (GLOD5)
GLOD5 belongs to the glyoxalase I family. Glyoxalase I catalyzes the isomerization of the hemithioacetal between a 2-oxoaldehyde and glutathione to form S-D -lactoylglutathione. The reaction mechanism probably involves a base-mediated proton transfer with an enediolate intermediate.Glyoxalase I (isomerase) is a member of the vicinal-oxygen-chelate superfamily which also includes as different enzymes as extradiol dioxygenase (oxidative cleavage of C-C bond), fosfomycin resistance protein (nucleophilic substitution), methylmalonyl-CoA epimerase and bleomycin resistance protein (no enzyme, sequestration). These proteins have in common that they share the same βαβββ-fold that provide a metal coordination site.
