C-Myc Binding Protein Associated Protein (MYCBPAP)

Using AMY1 (MYCBP) as bait in a yeast 2-hybrid screen of a human testis cDNA library, followed by a database search, Yukitake et al. (2002) identified a novel gene, which they designated AMAP1, encoding a deduced 947-amino acid protein. Northern blot analysis of various human tissues detected a major AMAP1 transcript of 3.5 kb and 2 minor bands of 4.8 and 7.7 kb in testis only, similar to the expression pattern of AMY1. RT-PCR of mouse testis showed that expression of Amap1 begins weakly at 3 weeks of age and is strong after the expression of spermatid, especially after 10 weeks of age, during which time AMY1 is also expressed.Immunofluorescence microscopy of HeLa cells expressing AMAP1 and AMY1 showed that both proteins colocalize in the cytoplasm, suggesting that AMAP1 is a bona fide AMY1-binding protein.
