Disrupted In Renal Carcinoma 3 (DIRC3)
By positional cloning and vectorette PCR to analyze genes disrupted by a t(2;3)(q35;q21) translocation in a family with renal cell cancer (RCC), followed by 3-prime RACE of normal kidney cDNA, Bodmer et al. (2003) cloned DIRC3. The predicted 3.0-kb transcript has a high density of stop codons in all 3 reading frames, making it likely that DIRC3 is a noncoding RNA.DIRC3 gene contains 12 exons and spans about 1 Mb.The TNP1 gene is about 400 kb proximal to DIRC3, and the TNS gene is about 300 kb distal to DIRC3. The breakpoints in both genes were intronic, and the fusion created a transcript where the first 2 exons of DIRC3 replaced exon 1 of HSPBAP1. The putative truncated HSPBAP1 protein retained its JmjC and HSP27 -interacting domains, which are associated with chromatin remodeling and stress response, respectively.
