Ras Like Family 12 (RASL12)

RASL12 is a member of the RERG branch of the Ras family of GTPases. RASL12 consists of the N-terminal core Ras domain and the C-terminal region of about 70 amino acid residues that is predicted to be unstructured. RASL12 lacks the C-terminal CAAX prenylation motif that anchors GTPases to membranes. This finding suggests that RASL12 may be mainly localized in cytoplasm. RASL12 may interact with transforming growth factor-beta receptor type I , activin receptor type I, bone morphogenetic protein receptor type IB and mothers against decapentaplegic homolog 1, 2, 3, and 4. the core domain of RASL12 bound to GDP in the presence of Endoproteinase Glu-C V8 and solved the crystal structure at 1.85Å. Superimposition with NRAS GTPase revealed that RASL12 adopted a conformation typical of Ras family GTPases in GDP-bound state.
