Rho Guanine Nucleotide Exchange Factor 16 (ARHGEF16)

Additional work identified the guanidine exchange factor ARHGEF16 (GEF16) as a novel binding partner for Tip-1 (manuscript in preparation). Rho GTPases play a fundamental role in numerous cellular processes that are initiated by extracellular stimuli that work through G protein coupled receptors. The encoded proteins -Rho guanine nucleotide exchange factors (Rhogef)- form complexes with G proteins and stimulate Rho-dependent signals.Arhgef 16 is thought to be involved in protein-protein and protein-lipid interactions. Family members share a common feature, a Dbl (DH) homology domain followed by a pleckstrin (PH) homology domain.Sixty-four novel structural analogues of Y27632 were synthesised and tested for their ability to persistently inhibit the transformation of NIH3T3 cells by Rho guanidine exchange factor 16 (ARHGEF16) or Ras.
