C2 Calcium Dependent Domain Containing Protein 4A (C2CD4A)

NLF1 and NLF2 share 75% identity in their C-terminal 291 residues, which contain extensive arginine-rich regions. NLF1 and its mouse ortholog share 38% amino acid homology. Lactacystin treatment of HMVECs abrogated the upregulation of NLF1 in response to IL1B, confirming the role of NF-kappa-B (NFKB) in regulating NLF1 expression. Luciferase reporter constructs demonstrated that putative NFKB binding sites in the NLF1 promoter were required for NLF1 upregulation in response to IL1B. Quantitative PCR showed NLF1 gene expression predominantly in endothelial cells; immunofluorescence analysis showed that transfected NLF1 localized to the nucleus in COS-7 cells, HeLa cells, and HMVECs. Higher resolution analysis showed that NLF1 was absent from areas of bright heterochromatin staining.
