By differential display PCR analysis to screen for genes expressed in stem cell factor (SCF)-deprived rat bone marrow myelomonocytic stem cells (MSC) that undergo rapid apoptosis, but not in differentiating or self-renewing MSCs, followed by RACE-PCR of rat and human brain cDNA libraries, Lucas et al. (2005) cloned rat and human DBNDD2, which they called HSMNP1. The deduced 161-amino acid human protein shares 86% sequence identity with rat Dbndd2, and both proteins contain a predicted N-glycosylation site, 3 consensus protein kinase C and casein kinase II phosphorylation sites, and a C-terminal PEST region. Genomic database analysis showed that expression was confined to mammals. Immunohistochemical analysis demonstrated cytoplasmic, vesicular localization during apoptotic induction.
