The rat ortholog was cloned and expressed in COS-7 cells, the homogenates of which showed transacylation activity, as did the purified enzyme. Unlike N-acyltransferase (NAT) from rat brain, which is highly Ca(+2)-dependent and membrane-bound, RLP1 was not stimulated by calcium and was detected mainly in the cytosolic fraction. RLP1 was also less specific that NAT as to the position of the fatty acyl chain. RT-PCR detected expression of RLP1 primarily in testis.rat RLP1 can transfer a fatty acyl chain from phosphatidylcholine (PC) to the amino group of phosphatidylethanolamine (PE), resulting in the formation of N-acylphosphatidylethanolamines (NAPEs), precursors of bioactive N-acylethanolamines such as the neurotransmitter endocannabinoid anandamide.
