ERR10 has 2 LxxLL motifs in its N-terminal half. Northern blot analysis detected ERR10 expression in thymus, prostate, testis, colon, and ovary, with minimal expression in spleen, small intestine, and peripheral blood leukocytes. Western blot analysis of ERR10-transfected 293T cells and in vitro translated ERR10 protein revealed a protein with an apparent molecular mass of 10 kD.The deduced 94-amino acid protein has a calculated molecular mass of 11 kD. It contains a transmembrane domain, a protein kinase C (PKC) phosphorylation site, and a myristoylation site. Northern blot analysis of 8 human tissues detected a 1.5-kb NAG7 transcript in skeletal muscle only. The N-terminal LxxLL motifs of ERR10 were required for inhibition of ER-alpha-mediated transcription.
