FAM84A, which shares 44% amino acid identity with FAM84B and 34.5% identity with C. elegans cuticle collagen-12 precursor. Semiquantitative RT-PCR and Western blot analysis confirmed upregulation of FAM84A mRNA and protein in colorectal carcinomas compared with normal colonic mucosa, as well as in bladder, pancreatic, and lung cancers. Northern blot analysis detected a 4.5-kb transcript in human testis only. Immunocytochemical studies of colorectal cancer cells localized FAM84A to the subplasma membrane region, limited to areas where cells did not have contact with neighboring cells. Western blot analysis detected a 32.5-kD species and a more abundant 33.5-kD species, and FAM84A was phosphorylated on ser38.
