Suppressor APC Domain Containing Protein 2 (SAPCD2)

p42.3 has an N-terminal EF-hand motif, a C-terminal coiled-coil domain, proline-rich regions that may function as protein-protein interaction domains, and numerous potential phosphorylation sites. Human and mouse p42.3 share 78% amino acid identity. RT-PCR, in situ hybridization, and immunocytochemical analyses detected p42.3 expression in embryonic tissues and gastric tumor tissues, but not in normal adult tissues. RT-PCR of cancer cell lines showed higher p42.3 expression in G1 and M phases than in S and G2 phases. RNA interference-mediated downregulation of p42.3 in a human gastric cancer cell line resulted in significant inhibition of proliferation and colony formation in vitro, development of a well-differentiated phenotype, and a significant reduction in tumorigenicity in nude mice.
