TRAF Interacting Protein With Forkhead Associated Domain Family, Member B (TIFAB)

TIFAB associates with TIFA and inhibits TIFA-mediated activation of NF-kappa-B (NFKB1). The deduced 161-amino acid protein shares 80% amino acid identity with its mouse homolog and contains 14 additional residues at the C terminus. Northern blot analysis of mouse tissues detected Tifab expression at high levels in spleen and at moderate levels in lung, thymus, and small intestine.mouse Tifab failed to activate NF-kappa-B when overexpressed in HEK293 cells, and they determined that TIFAB blocked TIFA induction of NF-kappa-B in a dose-dependent manner. Coimmunoprecipitation studies demonstrated that Tifab did not interact directly with Traf6. However, Tifab, Tifa, and Traf6 coimmunoprecipitated as a protein complex.
