Transforming Growth Factor-β1 stimulated clone-22 (TSC-22) is assumed to act as a negative growth regulator and tumor suppressor. TSC-22 belongs to a family of putative transcription factors encoded by four distinct loci in mammals. Possible redundancy among the members of the TSC-22/Dip/Bun protein family complicates a genetic analysis. In humans, two very similar long TSC22D2 isoforms are known, and mice have several TSC22D2 transcripts potentially coding for short TSC22D2 isoforms with distinct N-termini . TSC22D2 is involved in the osmotic stress response of mouse kidney cells. Since the potential redundancy among the various TSC-22/Dip/Bun family members renders a genetic analysis in mammals very difficult, it is important to assess the in vivo function of TSC-22 in a simpler model organism.
