Tetratricopeptide Repeat, Ankyrin Repeat And Coiled Coil Containing Protein 1 (TANC1)

Human and rat TANC1 share similarity with Drosophila rolling pebbles isoforms 6 and 7. RT-PCR of rat forebrain detected Tanc1 expression at postnatal day 1, with gradual increase in expression through 20 weeks. In situ hybridization showed widespread scattered distribution of Tanc1 in various brain regions, including intense expression in the hippocampal pyramidal cell layers and dentate granule cell layer, and moderate expression in the cerebellar cortex Purkinje cells and granule cells. Western blot analysis of cell-fractionated rat forebrain tissues detected a 200-kD Tanc1 protein in all membrane fractions examined, including the postsynaptic density and postsynaptic lipid raft fractions. No Tanc1 protein was detected in the soluble fraction. Immunohistochemical analysis localized rat Tanc1 protein to neuronal somas and dendrites.
