Triggering Receptor Expressed On Myeloid Cells Like Protein 2 (TREML2)

TREML2 is located in a gene cluster on chromosome 6 with the single Ig variable (IgV) domain activating receptors TREM1 and TREM2, but it has distinct structural and functional properties.The 321-amino acid protein has a calculated molecular mass of 35 kD. It contains an N-terminal leader sequence, followed by an IgV domain with 2 N-glycosylation sites, a transmembrane domain, and a short C-terminal cytoplasmic domain. The cytoplasmic domain has a tyr residue (Y315) within a potential endocytosis motif (YxxV) and another tyr residue (Y297) within a potent YxxC motif. Both tyr motifs are implicated in STAT3 recruitment and activation. The transmembrane domain of TLT2 lacks charged residues required for interaction with DAP12 (TYROBP).
