Zinc Finger Protein 300 (ZNF300)
ZNF300 contains C2H2 (or Kruppel)-type zinc finger motifs that tetrahedrally coordinate zinc ions and form a protein structure that can bind DNA and regulate transcription. The deduced 604-amino acid protein has a calculated molecular mass of 68.8 kD. ZNF300 contains an N-terminal Kruppel-associated box (KRAB) domain characteristic of transcriptional repressors, followed by 12 C2H2 zinc finger motifs that extend to the end of the protein. Northern blot analysis detected transcripts of about 4.5 and 3.1 kb expressed at highest levels in heart, skeletal muscle, and brain. Lower expression was detected in kidney, spleen, thymus, liver, and small intestine, but expression was not detected in other tissues examined. A transcript of about 7.0 kb was also detected in heart. Fluorescence-tagged ZNF300 showed nuclear localization in transfected COS-7 cells.
